Silencing it impairs tumor growth, making ROR1 a potential therapeutic target

A team of scientists at the University of California, San Diego Moores Cancer Center has identified a novel protein expressed by breast cancer cells but not normal adult tissues that could provide a new target for future anti-cancer drugs and treatments.

Led by Thomas J. Kipps, MD, PhD, Evelyn and Edwin Tasch Chair in Cancer Research and Interim Director of the UC San Diego Moores Cancer Center, the scientists found that the tumor cells of patients with breast cancer frequently express the Receptor-tyrosine-kinase-like Orphan Receptor 1, or ROR1. They found that silencing expression of ROR1 impaired the growth and survival of human breast cancer cells. The findings are published in the March 5 online issue of PLoS One.

ROR1 was first identified in the early 1990s and labeled an orphan receptor because its purpose was unknown. Subsequent work found that ROR1 is expressed at high levels during embryogenesis, during which time it plays an important role in regulating embryonic muscle and skeletal development. During fetal development, however, the expression of this protein is turned off. Normal cells and tissues in adults do not typically express ROR1.

Cancer cells, however, are a different matter.

"Cancer cells tend to acquire features of less differentiated cells," said Kipps, interim director of the UC San Diego Moores Cancer Center and a professor of medicine in the UC San Diego School of Medicine. "They often can be found to have features of embryonic cells."

In recent years, Kipps and others have become increasingly interested in the role of ROR1 plays in the growth of cancer and whether the protein might provide new options for stopping development of the disease. In 2008, for example, Kipps and colleagues reported that patients with leukemia treated with whole-cell vaccines could generate antibodies that reacted with their leukemia cells and the leukemia cells of other patients, but not with normal cells. They identified that such antibodies could target ROR1, accounting for the specificity of these antibodies in reacting with cancer cells. They identified another protein that could interact with ROR1 to stimulate the growth and/or survival of leukemia cells and that antibodies generated against ROR1 could block this function.

The discovery that ROR1 functions similarly in breast cancer heightens hopes. When the protein was silenced in human breast cancer cells, the cancer cells had slower rates of growth in the laboratory and in animal studies.

"There was a qualitative difference," said Kipps. "When ROR1 was knocked down, it took away some of the growth advantage enjoyed by cancer cells. Their capacity to survive also was impaired. This could affect the capacity of the cancer cells to survive treatment with other anti-cancer agents or generate tumors altogether."

The researchers report that expression levels of ROR1 correlate with the severity of at least some forms of breast cancer. The most aggressive cancers were the ones more likely to express ROR1. "That suggests ROR1 could be a good target for treating the most aggressive kinds of breast cancer, particularly the ones that lack expression of hormone receptors or the marker HER2/neu, which already can be targeted by monoclonal antibodies," Kipps said.

The discovery of ROR1's role in both blood and breast cancers also suggests it might have a similar function in other forms of cancer, a possibility Kipps said researchers will pursue.

National Day of Action for Education Rights | Fight Back!

Student protesters gather to read statement outside Governor Scott's door in Florida State Capitol.

Tallahassee, FL - On Thursday, March 1, the student movement in the United States took a collective step forward in the fight for higher education. Students for a Democratic Society (SDS), along with Occupy Education groups across the country and other progressive student organizations, held a nationwide day of action against crippling budget cuts, tuition hikes, and student debt.

Stephanie Taylor, an SDS organizer in Minneapolis-St. Paul, described this groundbreaking nationwide mobilization by saying, ?Nearly 20 SDS chapters nationwide protested on March 1st. While campuses focused on issues particular to their own areas, students throughout the country united with calling for an end to tuition hikes and to cut bloated administrators? salaries. Access to education is a right for everyone, not just for families who can afford it! We see the 99% on our campuses as the students, workers, and faculty who make the university run, and the 1% as the fat-cat administration on top who earn executive wages.?

In Chicago, Illinois, more than 100 students marched and rallied downtown at the Chase Bank Tower. A few students organized a short political theatre sketch, in which mock-representatives of Chase handed out fake diplomas in exchange for immense student debt. Afterwards, the students marched to DePaul University?s campus and held a sit-in to demand a meeting with President Rev. Dennis H. Holtschneider about rising tuition costs.

Looking south to Florida, over 50 students from Florida State University, Florida A&M University, Tallahassee Community College, and several local high schools gathered outside of the State Capitol to rally against budget cuts, tuition hikes, and exclusion of undocumented students from Florida?s public universities. After a few students gave militant speeches, they marched inside the Capitol, loudly chanting ?No ifs! No buts! No education cuts!? and ?They say cut back! We say fight back!?

Michael Sampson, an organizer with Progress Coalition and Uhuru FSU, said, ?Students from FSU, FAMU, TCC, and even high-schoolers came out to show that they are fighting back against attacks on education by Governor Rick Scott and the Florida Legislature.?

The group of students read a speech directly outside of Governor Scott?s door and proceeded to stage a sit-in on the fourth floor of the Capitol. Some parents who happened to be visiting the legislature that day then joined the students!

At the end of the rally, Sampson pointed out, ?Rick Scott was scared to come out of his office to meet us and hear our demands, but we won?t stop knocking at his door and next time we?ll bring hundreds. We?re not stopping!?

Further south in Gainesville, Florida, around 20 students from the University of Florida rallied outside of Tigert Hall, the Administration Building, to protest the University?s retention of Florida Senate President Mike Haridopolos as a professor making $75,000 annually. While taking a big paycheck from the University, Haridopolis has led the charge to slash higher education in Florida.

The University of Florida students hung an enormous banner across both doors of Tigert Hall reading ?Worst Professor Ever? in reference to Haridopolis. They then proceeded to fill out negative student evaluation forms, which denounced Haridopolis? attacks on students, workers, and undocumented families.

Marie Dino, an organizer in Gainesville Area SDS, said, ?Yesterday?s action was very powerful because our direct focus was on the source of these attacks on students. It?s time to get Haridopolis out of our campus and out of reach from our pockets!?

In Tampa Bay, SDS at the University of South Florida organized a walkout and a rally at the Marshall Student Center. Almost 200 students attended, and after several students gave militant speeches outside, the students forcibly occupied the Student Center and staged a sit-in, demanding an end to tuition hikes and budget cuts.

Corey Uhl, an organizer with Tampa Bay SDS, described the walkout as, ?a symbolic act that represents what classrooms will look like if these budget cuts and tuition hikes pass ? empty.? Uhl continued by saying, ?USF will no longer have a diverse student body if these cuts pass. That?s why we?re fighting back!?

When asked about SDS?s role in future nationwide actions, Taylor said, ?March 1st marked the third annual March day of protest for education rights and SDS was proud to be a major component of building for the day. This is just the beginning of a long spring of struggle and resistance to the 1% of our country devaluing education and making it more and more inaccessible for the average student.?

Florida Foreign Law Ban: Measure Banning Shariah, Other Foreign Law Progresses In Statehouse

WEST PALM BEACH, Fla. -- A measure to ban the use of foreign laws in domestic courtrooms is progressing in Florida's statehouse, one of dozens of similar efforts across the country that critics call an unwarranted campaign driven by fear of Muslims.

Forty such bills are being pursued in 24 states, according to a tally by the National Conference of State Legislatures, a movement opponents call a response to a made-up threat of Shariah law, the Islamic legal code that covers many areas of life. Backers of the bills say they fill a glaring hole in legal protections for Americans.

"There have been all sorts of wild accusations about what this bill does," said Sen. Alan Hays, R-Umatilla, who sponsored the Senate bill in Florida. "This is very clear, very simple: In American courts we need American laws and no other."

The Florida measure passed the House on Thursday 92-24. It awaits a full vote in the Senate.

If passed, Florida would join three other states ? Louisiana, Arizona and Tennessee ? in approving legislation curtailing the use of foreign laws. An Oklahoma ballot measure got 70 percent approval, but it goes a step further in specifically mentioning Sharia, the Islamic system of law. A federal court has blocked the measure's implementation until its constitutionality is determined.

The twin House and Senate bills in Florida make no mention of Shariah law or any other specific foreign system. The language of the legislation, in fact, seems innocuous, outlawing the use of foreign law only when it violates rights guaranteed by the U.S. Constitution, and only in certain domestic situations, such as divorces and child custody cases. It does not apply to businesses and says it shouldn't be construed to prohibit any religious organization from making judgments in "ecclesiastical matters."

But that's done little to quiet critics who see such legislation as right-wing fear mongering.

"It's a waste of time and irrelevant legislation," said Nezar Hamze, head of the Miami chapter of the Council on American-Islamic Relations. "But the motive behind it is very troubling."

The most fervently outspoken supporters of such bills caution Shariah law could begin to spread outside of Muslim countries in a slow-speed Islamic takeover of the world. Others, seeking to appeal to the masses, say not outlawing Shariah jeopardizes the rights of American women.

Though Shariah law was an unrecognizable term to nearly every American just a few years ago, it has become much more mainstream. Dangers of Shariah have been aired on the campaign trail, in tea party rallies and on cable news.

One of the most persistent voices on the issue is David Yerushalmi, a Brooklyn lawyer who drafted model legislation on the foreign law issue and who has waged a quiet campaign to ensure Shariah is outlawed in the U.S.

Yerushalmi's views have made him a lightning rod; he even declines to say where in Florida he lived as a child because he has family that still calls it home and he says he fears for their safety. He disputes characterizations of him as a bigot.

Yerushalmi calls Shariah "an offensive foreign law" but he says even if critics are right, and that he and other proponents of such legislation are acting on prejudice, legislatures have nothing to lose by outlawing it.

"If you're right and Shariah is everything that is good and noble and doesn't have all the ugly things that we understand it to have," he said, then such legislation simply will have no effect on the public. He notes people found to have committed adultery can be stoned under Shariah law.

The Florida bills include passages from Yerushalmi's model legislation, which was written for a group called the American Public Policy Alliance. The leader of that organization, Stephen Gele, says there are egregious court cases that have shown Shariah is a threat in the U.S., with foreign judgments on divorces and child custody allowed to stand.

"It's probably a small percentage of a court's docket, but certainly if you're the woman who lost her child to Pakistan, it's important to you," he said, citing a case in which he said a mother lost custody because of a foreign ruling.

An estimated 1.3 million Americans identify as Muslim, according to the 2008 American Religious Identification Survey, a respected count published by Trinity College in Hartford, Conn. That amounts to fewer than 1 percent of the population nationally. Florida has a similar share.

If passed, Hamze said, Florida's legislation would have virtually no effect because he says he's not aware of the issue ever coming up in a state courtroom. He believes the message, however, would be a harmful one.

Others have come to the side of Muslim advocates, including the Anti-Defamation League, a prominent defender of Jewish causes. Andrew Rosenkranz, regional director for the ADL in Florida, issued a statement calling the bills unnecessary and saying they could jeopardize agreements reached in religious settings such as a Jewish tribunal.

"The alleged threat of Islamic, other religious or foreign law to Florida's court system is completely illusory, and the Senate's consideration of this measure is an unwise use of resources," Rosenkranz said. "At best, this bill is wholly redundant as the Florida and U.S. constitutions already prohibit the unconstitutional application of foreign law in the courts."

Hays said those who oppose the bill are mistaken about its intentions. He said he wasn't aware what led to the legislation's introduction ? another version of it failed last year ? or who might have had a role in crafting its language, which he said was ultimately put together by the House sponsor, Rep. Larry Metz, R-Yalaha. Metz did not return a call seeking comment.

"I don't care where the law comes from or who the originators of the law are," Hayes said. He said those with concerns need to do just one thing to ease their fears: "Read the bill."

Stress Free Home Entertaining | Living better at 50 | Online ...

By Jane Heacock ?

Some people are born great organizers others are not. Holiday entertaining, birthday celebrations and dinner parties can be stress free with a little bit of planning. Recently I organized a 60th birthday party. The planning of this party was put together in just four weeks. Here are my party planning tips that can be applied the next time you are hosting a party.

Invitations

For this particular 60th birthday party I sent out the invitations four weeks in advance. If you are inviting out of state family or friends be sure to give them more than the four weeks notice. There are many options for invitations depending on the occasion. Evites is a popular way to get the word out about your party and a great way to keep track of the number of people you can expect. Send an email reminder when the date is getting closer for those that have not sent an RSVP.

Planning a Family Gathering

Depending on the type of party you are having attendees sometimes offer to bring appetizers, wine or their favorite party food. For our family gatherings the host puts a menu together and sends an email out listing what is needed. Family members select what they would like to bring and let me know in advance. If I am hosting I supply the main part of the meal as well as all the condiments, drinks and party supplies. My family then brings appetizers, side dishes and some of the desserts. This gives me time to get the house in order and set everything up before the party day. Not having to prepare all the food takes the stress off my shoulders. I usually start setting up tables, set up party supplies and have the house cleaned at least three days before the party.

Formal Parties

For business parties, holiday socials, a theme party or a special event party I start planning at least two months ahead. I start organizing my lists earlier because I order some of food from local specialty shops. If you will be ordering a theme cake it is good to put your thoughts together earlier rather than later, as the party date comes closer you will have other details to attend to. Start a detailed computerized list that way you can update each area of your party planning as time goes along. List the pricing for each area of your party planning to stay within your budget.

Finally enlisting some assistance for set up and clean up is a smart way to keep your party planning stress free-?Arrange to have your home cleaned and hire a personal assistant company, this gives you time to concentrate on other areas of your party organizing. Have your personal assistant pick up your decorations, party platters, cake etc. When you are at the two day mark before your party you will have lots of other details to attend to. Do yourself a favor and enlist some assistance!

Last but not least, give yourself at least one hour before your party to sit back and relax. You deserve it!

Are you ready for the best Paleo crock pot recipe you?ve ever tasted? Brace yourself? You?re about to experience the fruit of Sam?s slow-cooking kitchen mastery: beef burgundy with sweet potatoes.

As a man with a short attention span and a penchant for improvisation, I tend to put up quick recipes that are best-suited for small crowds and?accelerated?appetites. But not Sam. Her beef burgundy is a slow-cooking show-stopper capable of making entire auditoriums salivate. And prepare to enjoy your kitchen time. If you?re anything like Sam, you?ll be singing at the top of your lungs, brandishing fresh herbs, and breaking out that special bottle of Chilean cab sauv.

And without further adieu, I?ll turn it over to Sam.

Hello. I'm Sam.

?Since even people who aren?t on any kind of diet can savor these intense flavors, beef burgundy is a true crowd-pleaser.

Cook it in large quantities to feed the entire family or to wow guests; it?s especially good for piquing the interest of those that are on the fence about this whole new lifestyle you?re rolling with.

It also makes outstanding leftovers (I never get around to re-heating them, it smells so good cold I just yummy on down?).?

Shopping and Preparation Tip:

Grab your ingredients the day before, since you?re soaking all the lovely beef and veggies overnight in wine and burgundy. That?s right ? now you can drink your wine and eat it, too!

Oh, and if you don?t have a slow-cooker, you can put the whole dish in an oven-safe, large pot with a tight-fitting lid and cook on low heat. It cooks on the stove top as well, but that means at least 3 hours in front of the stove. Nah, that?s cool. I?d rather not.

Here?s the Beef:

  • 2 to 3 lbs shoulder or roast beef (more beef is always better, and the tougher the cut of meat the better it holds up in the cooking)
  • 1 Medium sweet onion, chopped
  • 1 cup sliced carrots
  • 1 cup sliced celery
  • 4 cloves of garlic, chopped or minced
  • 1 sprig or 1 tbsp fresh rosemary (some prefer sage, I go either way)
  • 1 sprig or 1 tbsp fresh thyme
  • 1 tps crushed black pepper
  • ? cup peeled shallots (optional)
  • 1 tsp cloves (optional)
  • 1 bottle of dry to medium red wine (I tend to go for less pricey bottles when I?m cooking with them. But then, I also tend to pour myself a half a glass to keep the cooking process robust!)
  • 1 cup of burgundy wine (I sometimes use grappa when I?ve got it on hand)

The Toppings

  • 1 cup sliced mushrooms
  • ? cup chopped bacon
  • 1 cup pearl onions (white or red, both are tasty!)

The Unconventional Roux

  • 4 tbsp butter
  • 4 tbsp tapioca flour (this gluten-free flour doesn?t affect the flavor of the dish)

The Sweet Potatoes

  • 3 to 4 sweet potatoes
  • 4+ tbsp real butter or ghee
  • Thyme and pepper (optional)
  • ? cup cream

So here?s the flow for this dish: get your ingredients the day before. Chop and prep the first set of meat and vegetables, then soak overnight. In the morning, you?ll be browning the meat and tossing everything into a slow cooker. It takes several hours on low heat to make all that meat melt in the mouth. That night, with a few more minutes of prep, you?ll have a completed meal.

This is a show-off meal, and although the total time to make it is a bit much, that time is disbursed into three little pockets of time spread out over a day and a half.

The Night Before

Cube the beef, about 1 ? inches on each side. This makes them more manageable while cooking, and they?ll cook down to closer-to-bite-sized. Then they?ll just fall apart from being tender anyway.

Toss the cubed beef into a large bowl. Add the onion, carrots, celery, garlic, rosemary or sage, thyme and black pepper (also add the optional cloves and shallots if you?ve got ?em). Pour the wine and the burgundy over it, stir, and cover (I love wine, I love wine, winey-wine-wine, here it goes down, down into the crock pot!). Put the whole shebang in the fridge overnight to soak.

The Morning of the Feast

In the morning, marvel at the amazing smell, and let the anticipation build. Here?s the secret to the intense savory flavor: you?re going to fish out the cubes of beef and brown them in a skillet with a drizzle of olive oil. Brown each side (this is where the larger chunks come in handy) to lock in those meaty juices, then toss into the crock pot. At this point, go ahead and turn the crock pot on.

Here?s another spot you can choose to go all out or save yourself some time. You can fish out the veggies and brown them, too, or you can just take the entire bowl of wine and veggies and pour it into the skillet (or another pot while you?re browning that meat) and get it nice and hot. Once the veggies and wine sauce are hot, pour into the crock pot and cover. Let it hang out there until you?re ready to start making dinner that night.

Yes, it?s going to make the entire house smell amazing all day long.

Salivating yet?

Showtime

About a half hour before you intend to serve dinner, cook the bacon to a nice crisp. Toss it into the slow cooker. Then brown the mushrooms and pearl onions in the bacon fat. Toss them into the crock pot as well and cover it up.

Make a tapioca roux by heating the butter, then adding the tapioca flour. Cook until lightly brown, then add to the slow cooker and stir everything well.

Now for the sweet potatoes. I often cheat and make these ahead of time, then re-heat them in the oven (or, yes, even the microwave). You can either boil the cubes until tender, then add the butter and spices when you mash them. Or, if you?re itching to really impress, roast the cubes in the oven with butter and spices at 350 degrees for about 20 minutes. Mash the potatoes up well and whip with the cream. Add more butter to desired taste and consistency.

Dollop the creamed sweet potatoes onto each plate and use a spoon to press down the centers to make a little bowl. Ladle an overflowing heap of the beef burgundy into your sweet potato bowl. When those assembled at the table start ooh-ing and ahh-ing, feel free to jibe a bit (?Mmmm. This looks good. I mean, really good. Hey, everyone! Come and see how good this looks!?).

Then sit and sip with the rest of the bottle of burgundy, enjoying the way the beef falls apart in the mouth and the sauce melts you to your toes. You are now a royal cook, rocking food that helps you burn fat, build muscle, and become the envy of every kitchen.

Time Savers

If you don?t think you?ll have time for chopping the veggies, grabbing some pre-chopped veggies from the co-op or grocery store will save a bit of time.

If you suddenly don?t have time in the morning, you can omit browning the meat. Warning: this is really going to cut down on flavor and meat consistency, so only do this in dire circumstances.

Cook the sweet potatoes in the microwave. Not nearly as sexy as roasting or even boiling, but it?ll get the job done. Use a fork to punch several holes all over them (if they aren?t already diced), rub them with the butter and put them in an open plastic storage bag. It might take 10 to 15 minutes in the microwave to cook them whole, about 5 to 10 minutes to cook the cubes. All this depends on microwave strength, so keep an eye on them. (Note, we don?t like using microwaves, so avoid this if you can.)

Writing this post makes me hungry...

Carb Savers

If you?ve decided you need to keep the carbs super low, you can eat this as an intense stew. Omit the roux and the sweet potato part of the recipe altogether and add hot water to thin the beef concoction. Omit the carrots as well. You can sub in other vegetables (celery root and spaghetti squash will give it a more intense consistency) or just up the amounts of the other veggies already in the recipe to make up for the loss.

Pairings

Did I mention the wine? Ah, yes. I did. But it bears repeating. I tend towards Cabernet with beef, but if you?ve already opened the bottle of Burgundy?

Anything else? Not really, though a side salad of baby spinach with olive oil, salt and pepper, grape tomatoes and feta cheese can be whipped up quick while you wait for the dish to come together. Also adds a nice bit of color to the plate.

You won?t have room for desert at the end, but if by some strange twist of fate you do, some camembert or brie cheese pressed with sliced almonds tastes lovely with the wine.

Stay classy, folks.
