Using the Canadian Light Source synchrotron, researchers from the University of British Columbia have revealed, for the first time, one of the molecular mechanisms that regulates the beating of heart cells by controlling the movement of sodium in out of the cells and what calcium has to do with it. The findings, published February 14 in the Proceedings of the National Academy of Sciences, sheds new light on this crucial physiological process while revealing the root cause and possible treatment targets of two potentially life-threatening cardiac arrhythmia conditions.

The contraction and relaxation of heart muscle cells depend on minute but finely regulated electrical impulses that are created when charged atoms or ions of metals such as sodium, potassium and calcium pass through complex molecular channels inside and between cells. Irregular heartbeats, referred to medically as arrhythmias, can happen when these channels leak or otherwise malfunction. Professors Filip van Petegem and Christopher Ahern, members of UBC's Cardiovascular Research Group, used the CLS to determine the molecular structure of a part of the channel that controls the flow of sodium to cells in the heart, as well as in other electrically-excitable cells such as in the nervous system.

"The heart is an electrical organ that depends on precise electrical signals to contract [and pump blood]" explains van Petegem. "It is crucial for heart rate that the signalling, controlled by the movement of sodium, be exact. So the entry of sodium into the cell is tightly regulated."

The sodium channel that passes through the outer membrane of heart cells is actually a huge, intertwined four-part molecule. The teams of Van Petegem and Ahern chose a section of the molecule that appeared to regulate the closing of the channel by forming a plug, thus stopping sodium from getting through.

The researchers were surprised to discover that a protein called calmodulin binds to the sodium channel, keeping it open by preventing the plug from forming. Calcium ions, in turn, regulate the connection between the protein and the channel: calcium ions cause the protein to hook up to the channel, keeping it open and letting sodium through.

Problems occur with the system when genetic mutations change the shape of the channel at the site where the protein binds, affecting how well the channel can open and close. The result the flow of sodium into the muscle cells is disrupted and the heart does not beat regularly.

The scientists have been able to identify mutations in the site that lead to two different kinds of heart arrhythmia: Brugada Syndrome and Long Q-T type 3, so-called from the tell-tale trace doctors see on the ECG of patients suffering from the problem. Brugada syndrome is considered to be caused by not enough sodium getting into cells, while long Q-T is the result of too much sodium.

LIMA (Reuters) ? The most important leader of Peru's leftist Shining Path insurgency has been found dead by security forces after being shot in a remote jungle rife with drug trafficking, President Ollanta Humala said on Sunday in his first major victory against what remains of the rebel group.

Artemio, the nom de guerre of Florindo Eleuterio Flores, headed a remnant group of guerrillas that went into the cocaine trade after the founders of the Maoist rebels were imprisoned during a bloody war against the state in the 1990s. China imports slump, raising demand concerns (Reuters)

BEIJING (Reuters) ? China's imports in January fell the most since the depths of the global financial crisis, raising concerns that demand may be weaker than previously thought even allowing for Lunar New Year factory shutdowns.

Imports sank 15.3 percent in January versus January 2011 -- the lowest since August 2009 -- while exports fell 0.5 percent over the same period, the worst showing since November 2009, customs data showed on Friday.

While raising worries about the resilience of domestic demand, which has shielded the world's second-largest economy from slackening exports, it also raises alarm bells about China's ability to support a frail global economy.

"A fall of over 15 percent in January cannot be entirely explained by the Lunar calendar, and adds weight to the view that economic output is slower than headline indicators might suggest," said Ren Xianfeng, an economist at IHS Global in Beijing.

Still, Lunar New Year distortions will make policymakers wary of any hasty reaction. Analysts expect them to assess January and February data combined before deciding whether the current policy of gentle easing should be intensified.

The big imports drop combined with a smaller exports drop left China with a trade surplus of $27.3 billion in January, its biggest in six months and confounding expectations of a further narrowing.

Exports to the European Union, China's top export market, fell 3.2 percent in January from a year earlier, the first decline since February last year, the data shows.

Exports to the United States rose 5.5 percent in January from a year earlier, slowing from December's 11.9 percent rise and marking the weakest pace since February last year.

Other figures on Friday showed China's current account surplus shrank in 2011, offering Beijing fresh evidence to show critics of its currency policy that it is relying less on external demand.

Chinese leaders can point to the figures next week at a summit with European Union officials in Beijing, as can Vice-President Xi Jinping, widely expected to be China's next leader, who visits Washington on Tuesday.

The Australian dollar edged lower after the trade data as investors worried it pointed to weaker China demand for the country's commodities.

Still, analysts cautioned against reading too much into a single month of data skewed heavily by the seasonal distortion of the week-long Lunar New Year holiday, which fell in January this year and in February last year. Factories typically shut or run at half speed during this period.

That left economists reading between the lines with the wide range of their forecasts a clear sign of uncertainty.

"The drop in both January exports and imports is not drastic enough to trigger any aggressive move in monetary policy," Sun Junwei, economist at HSBC Global Research in Beijing, said.

"The decision makers may probably look at more indicators for February and other data such as foreign exchange purchase positions to gauge the policy stance."

Analysts at Bank of America/Merrill Lynch said in a client note that making swift adjustments for actual days worked showed the data in a different light.

Under such an adjustment, exports may have grown 28.7 percent from a year earlier and imports may have risen 10 percent given that there were just 17 working days in January compared to 22 working days a year ago.

LIMITED ROOM FOR EASING

China's economic expansion struck a 2- year low of 8.9 percent in the last three months of 2011, extending a steady slowdown that had prompted the government in the autumn to switch policy settings to support growth. It has gently eased monetary and fiscal conditions since.

The central bank cut the bank reserve ratio in late November for the first time in three years and is expected to reduce it further to help offset an expected decline in capital inflows. A fall in these inflows hinder money supply growth and so economic growth, analysts say.

The market had anticipated a cut in banks' reserve requirement ratio (RRR) ahead of the Lunar New Year. But the People's Bank of China, the central bank, has opted instead to sue open market operations to inject short-term liquidity into the financial system.

Many analysts reckon a cut in the RRR may now be set back until March after data on Thursday showed annual inflation spiked to a consensus-busting 4.5 percent in January.

"We do not believe there will be imminent direct policy implications from this data because policy makers understand January data are all highly distorted across the board.

"Taken at face value today's trade data should push policy makers to loosen policy further. This may well happen if February year-on-year exports data is still as low but we believe there will be a large rebound to double-digit levels," Yu Song, China economist at Goldman Sachs, said in a note to clients.

"At the same time, the pressures to loosen from trade data are offset by yesterday's CPI data, which surprised on the upside, which taken at face value would point to a need to tighten policy."

Few analysts believe the central bank will cut interest rates this year, with annual inflation staying stubbornly higher than the one-year deposit rate of 3.5 percent.

STABLE YUAN

The slope of China's economic slowdown has been shallow enough for the consensus to emerge that a hard economic landing will be avoided, even though many private-sector economists forecast that 2012 will see the slowest pace of expansion in a decade.

The first quarter of 2012 is widely expected to mark the bottom of China's economic downswing. Signs from the most recent purchasing managers index survey showed a slight expansion of the factory sector in January.

China's minister of commerce, Chen Deming, had flagged that January's numbers would not look pretty in a statement released on Thursday, adding that a stable yuan currency was needed to help Chinese exporters.

However, the closely managed yuan rose to a record intraday high of 6.2884 per dollar on Friday, which traders said would be designed to appease long-festering concerns in the West that China holds down the value of the currency to boost exports.

Xi is likely to hear calls in the United States next week that Beijing allow the currency to rise at a faster rate. The EU is also likely to make such calls when it meets China's leaders.

However, a Reuters poll this month suggested such hopes may be dashed. It showed that China is expected to slow the pace of yuan gains to help exporters cope with the slowdown in global demand growth.

The yuan is likely to gain 2.8 percent over the next 12 months, slower than the 4.7 percent rise in 2011.

China's State Administration of Foreign Exchange (SAFE) -- guardian of the country's $3.18 trillion of official reserves that have become the world's biggest largely on the strength of Chinese exports -- said the country's current account surplus would fall sharply in 2012.

SAFE said though that the surplus, which narrowed in 2011 to 2.7 percent of GDP from 5.1 percent in 2010, would remain despite the risk of external shocks to demand and capital flows.

Feb 10 (Reuters) ? Wegelin & Co, the oldest Swiss
private bank, was declared a fugitive after failing to show up
in a U.S. court to answer a criminal charge that it conspired to
help wealthy Americans evade taxes.

At a hearing in Manhattan federal court, U.S. District Judge
Jed Rakoff on Friday suggested that U.S. prosecutors enlist the
help of diplomatic authorities, including perhaps the State
Department, to advance the case.

The indictment of Wegelin, which was founded in 1741, was
the first in which the United States accused a foreign bank,
rather than individuals, of helping Americans commit tax fraud.

Wegelin was accused of helping clients hide more than $1.2
billion in offshore bank accounts.

?Occasionally in these situations, progress is made through
diplomatic channels,? Rakoff told Assistant U.S. Attorney Daniel
Levy at the hearing. ?Unlike an individual, arresting a company
is somewhat difficult, other than in science fiction.?

Rakoff spoke after Levy said ?we have no proposal? for how
to get Wegelin to formally answer the charge.

Prosecutors unveiled the charge on Feb. 2, one month after
bringing conspiracy charges against three bankers in its Zurich
branch: Michael Berlinka, Urs Frei and Roger Keller.

According to prosecutors, more than 100 U.S. taxpayers
conspired with the defendants and other unnamed co-conspirators
between 2002 and 2011 to hide money from the Internal Revenue
Service.

The government also seized more than $16 million from an
account that Wegelin held in Stamford, Connecticut with the
Swiss bank UBS AG.

Wegelin has no branches outside Switzerland, and followed
the common industry practice of using correspondent banking
services to handle money for U.S. clients.

In 2009, UBS paid $780 million and entered a deferred
prosecution agreement with the Justice Department to resolve
allegations that it helped Americans evade taxes.

Wegelin effectively broke up last month by selling its
non-U.S. operations to the Swiss bank Raiffeisen.

It also moved most of its workers, clients and 21 billion
Swiss francs (US$22.9 billion) of assets to Notenstein
Privatbank, set up specifically for the break-up.

No further proceedings are scheduled in the Wegelin criminal
case.

The case is U.S. v. Wegelin & Co et al, U.S. District Court,
Southern District of New York, No. 12-cr-00002.

Cambridge, MA - Move over, coffee and Red Bull. A Harvard professor thinks the next big thing will be people inhaling their caffeine from a lipstick-sized tube. Critics say the novel product is not without its risks.

The product, called AeroShot, went on the market late last month in Massachusetts and New York, and is also available in France. A single unit costs $2.99 at convenience, mom-and-pop, liquor and online stores.

Biomedical engineering professor David Edwards said AeroShot is safe and does not contain common additives, like taurine, used to amplify the caffeine effect in common energy drinks. Each grey-and-yellow plastic canister contains 100 milligrams of caffeine powder, about the amount in a large cup of coffee, plus B vitamins.

But Democratic U.S. Sen. Charles Schumer of New York wants the U.S. Food and Drug Administration to review AeroShot, saying he fears it will be used as a club drug so that young people can drink until they drop. Schumer?s national press secretary did not immediately respond to calls for comment.

FDA spokeswoman Siobhan DeLancey declined to comment, saying the agency will respond directly to Schumer on the matter.

Edwards said Schumer?s comments are understandable in the context of developments over the last few years, when students looking for a quick and cheap buzz began consuming caffeine-packed alcoholic drinks they dubbed ?blackout in a can? because of their potency. But he said AeroShot is not targeting anyone under 18 and it safely delivers caffeine into the mouth, just like coffee.

?Even with coffee ? if you look at the reaction in Europe to coffee when it first appeared ? there was quite a bit of hysteria,? he said. ?So anything new, there?s always some knee-jerk reaction that makes us believe ?Well, maybe it?s not safe.??

Once a user shoots a puff of calorie-free AeroShot into his or her mouth, the lemon-lime powder begins dissolving almost instantly. Each single-use container has up to six puffs.

?The act of putting it in your mouth is the act of breathing ? so it?s sort of surprising and often people the first time they take the AeroShot, they laugh ... that it?s kind of a funny way of putting food in your mouth,? said Edwards, who also came up with a breathable chocolate product a few years back.

Dr. Lisa Ganjhu, a gastroenterologist and internal medicine doctor at New York-based St. Luke?s-Roosevelt Hospital, said people need to be aware of how much caffeine they are ingesting.

?You want those 10 cups of coffee, it will probably take you a couple hours to get through all that coffee with all that volume that you are drinking,? Ganjhu said. ?With these inhale caffeine canisters you can get that in 10 of those little canisters ? so you just puff away and you could be getting all of that within the hour.?

Even the product packaging warns people not to consume more than three AeroShots per day.

Northeastern University students who sampled the product recently gave it mixed reviews.

?This tastes really good and I think it rocks,? student Zack Huang said after puffing onto a free sample before rushing to join a group of friends who were walking away from campus.

Still, one student was not happy with the taste, echoing sentiment expressed online by some consumers.

People elsewhere vowed they would never give up their morning coffee.

?I want to brew it, I want to stir it and I want to drink it slowly as I absorb the caffeine,? said longtime coffee fan Mark Alexander.

The makers of AeroShot appear to be aware of that sentiment, declaring that the product isn?t about switching away from coffee, but rather making it easier for people with active lifestyles to get their caffeine fix.

?AeroShot can be used in a variety of settings inconvenient for liquids, such as when you study in the library, board an airplane or get into the car for a long drive,? they say in the section dedicated to frequently asked questions on their website. ?It?s easy to take AeroShot with you when you go biking, skiing, curling, or any other activity that consumes energy.?

AeroShot, manufactured in France and the flagship product of Cambridge-based Breathable Foods Inc., is the product of a conversation that Edwards had with celebrity French chef Thierry Marks over lunch in the summer of 2007.

?We were discussing what interesting culinary art experiments we might do together and I had the idea that we might breathe foods since I?ve done a lot of work over the last 10 or 15 years on medical aerosols,? Edwards said.

The first venture Edwards worked on with Harvard students was the breathable chocolate, called Le Whif. Now he?s preparing to promote a product called Le Whaf, which involves putting food and drinks in futuristic-looking glass bowls and turning them into low-calorie clouds of flavor.
